N-desmethyl-sildenafil in terms of rate and extent of bioavailability.
There was a washout interval of ≥7 days between the two administrations of the investigational medicinal products. Blood samples for pharmacokinetic analysis were collected up to 24 h post-dosing. The primary objective was to compare the rate (peak plasma concentration; Cmax) and extent (area under the curve [AUC] from administration to last observed concentration time; AUC0–t) of sildenafil absorption after single-dose administration of test and reference. Secondary endpoints were observed to describe the plasma pharmacokinetic profiles of sildenafil and its metabolite N-desmethyl-sildenafil relative bioavailability and safety profile after single-dose administration. The mean sildenafil and N-desmethyl-sildenafil plasma concentration–time profiles up to 24 h after single-dose administration of sildenafil 100 mg orodispersible film and film-coated tablet were nearly superimposable. Adverse events occurred at similar rates for
the two formulations and were of mild-to-moderate severity.

My doctor advised me to consider either radiotherapy or removal of the prostate. Since the cancer is very small, I plan to first try managing it with a special diet. However, I believe this issue is unrelated to my erectile problems. There are no further issues or medications involved. Thank you for providing the additional information promptly.

“The FDA approval of Vybrique is genuinely exciting and adds a fresh option to the ED treatment space. What stands out is how easy it is for patients to use: it can be taken with or without food, there’s no need for water, it dissolves in the mouth, and it works faster than traditional sildenafil pills. Those details matter, especially when spontaneity and convenience are a big part of sexual health,” she explained. “My patients are always asking about new and innovative therapies. I’m looking forward to seeing how Vybrique performs in the real world and how it fits into everyday clinical practice as another way to personalize care.” IBSA USA announces FDA approval of VYBRIQUE™, the first and only oral film to treat men with erectile dysfunction.

The pharmacokinetics of the sildenafil 100 mg orodispersible film (IBSA) was compared to that of the conventional marketed 100 mg film-coated tablet (Viagra®) after single-dose administration to 53 healthy male volunteers (aged 18–51 years) in a randomized, open, two-way crossover bioequivalence study. Each subject received a single oral dose of 100 mg of sildenafil as test or reference formulation administered under fasting conditions at each of the two study periods according to a randomized crossover design. Keywords: sildenafil, pharmacokinetics, bioequivalence, orodispersible film,
Patients younger than 65 years of age received a starting dose of 50 mg, and patients 65 years and older received a starting dose of 25 mg. The trial was conducted as a flexible-dose study, meaning that the starting dosage could be increased or decreased. Overall, the study met its co-primary end points, with VYBRIQUE demonstrating superior improvements in sexual function vs placebo. Pharmacokinetic data also showed that peak plasma concentrations were reached between 30 and 300 minutes under fasting conditions. The median time to peak plasma concentrations was 80 minutes.
“While awareness and open discussion around ED have grown, it’s still a sensitive topic where many men value discretion,” added Nicholas Hart, IBSA USA CEO, in the news release.1 “The FDA approval of VYBRIQUE provides men experiencing ED with a novel treatment option that helps meet the evolving needs of patients today.” The sildenafil oral film is expected to be commercially available in the US in March 2026. IBSA also reported that direct home delivery of VYBRIQUE will be available in all states, “providing patients a convenient fulfillment option.” Commenting on the approval to Urology Times, Justin Dubin, MD, said, “As men continue to face stigma around men’s health – especially erectile dysfunction – it’s encouraging to see more discreet, patient-centered treatment options become available. Fast-acting PDE5 inhibitor troches with sublingual or buccal absorption have been available through compounding pharmacies for years, but they’ve lacked FDA approval. With the FDA now approving a fast-acting, easy-to-use ED therapy, this represents a meaningful step forward in patient care and reinforces a commitment to privacy, discretion, and efficiency in both treatment and medication delivery.” Dubin is director of Men’s Sexual Health at Baptist Health Miami Cancer Institute in Florida. Amy Pearlman, MD, a board-certified urologist with expertise sildenafil citrate tables in sexual, hormonal, and genitourinary health from South Florida, also spoke to the convenience of the oral film and its implications for patient care. VYBRIQUE is a sildenafil oral film formulated for discreet, water-free administration, aiming to sildenafil citrate drug reduce use barriers for patients for whom privacy and convenience influence ED treatment adherence. A phase 3, 12-week, randomized placebo-controlled trial (n=475) met co-primary endpoints, demonstrating statistically superior sexual function outcomes versus placebo with multiple dose strengths. The sildenafil oral film is expected to be commercially available in the US in March 2026.
The FDA has approved the first sildenafil oral film, VYBRIQUE, for men aged 18 years and older with erectile dysfunction (ED), IBSA USA announced in a news release.1 According to IBSA, VYBRIQUE is “a discreet, easy-to-use, single-dose film that dissolves on the tongue without the need for water or other liquids.” The oral film can be taken anywhere from 30 minutes to 4 hours before sexual activity and can be taken with or without food. “Beyond age-related factors, research2 shows that anxiety, depression, and stress can contribute to ED,” said Ralph M. Zagha, MD, Precision Clinical Research, in the news release.1 “Providing a treatment option that is discreet and easy to administer supports men in managing ED in a way that feels approachable and respectful of their sexual health.” The approval of the oral film is backed by findings from a phase 3 randomized trial (NCT05490680), which demonstrated that VYBRIQUE provided superior improvements in sexual function vs placebo. In total, the 12-week study enrolled 475 adult men with ED across clinical trial sites in the US.3 Participants were randomly assigned to receive VYBRIQUE or placebo. Doses of VYBRIQUE were administered at 25 mg, 50 mg, 75 mg, and 100 mg.
